Nano-Traps are ideal for fast, reliable and efficient one-step immunoprecipitations

Nano-Traps pull down the proteins of interest (POI) and their interacting factors from cell extracts or organelles. Nano-Traps consist of a VHH antibody, also termed a “nanobody”, coupled to an immobilising matrix (agarose beads, magnetic agarose beads, magnetic particles1 or multiwell plates1).

: All Nano-Traps are available coupled to agarose beads and in 96-multiwell plate formats. Selected Nano-Traps are also available coupled to magnetic agarose beads or magnetic particles.

What are VHH antibodies?

Next to conventional IgG antibodies, which are composed of two heavy chains and two light chains, the immune repertoire of camelids such as dromedaries, llamas and alpacas contains so-called heavy chain only IgGs. These antibodies lack the CH1 domain of the heavy chain and are devoid of any light chain. Therefore, the variable domain of these heavy chain only IgGs, called VHH or Nanobody, represents a stand-alone antigen binding entity and can be produced as a single domain antibody fragment (sdAb) of only 15kDa.


What makes ChromoTek’s technology special?

For more than a decade, ChromoTek has generated highly specific and high affinity VHH antibodies for various applications. By combining their proprietary immunisation scheme with phage display, ChromoTek are capable to create recombinant sdAbs with tailor-made properties. This technique provides the best of both worlds: Highly specific antibodies that have undergone affinity maturation in vivo, but that are produced recombinantly with known sequence and without any batch-to-batch variations. Typically, affinities are in the picomolar to single-digit nanomolar range. Subsequently, the VHH domains can be further engineered or adapted, depending on the requirements of a specific application.

The GFP-Trap® and RFP-Trap®


Description: The ChromoTek GFP-Trap® for one-step immunoprecipitation (IP) of GFP-fusion proteins. GFP-Trap is an anti-GFP VHH coupled to agarose or magnetic agarose beads. The anti-GFP VHH is also known as GFP-Binding-Protein, GFP Nanobody or anti-GFP single domain antibody (anti-GFP sdAb). ChromoTek GFP-Trap is referenced in more than 1,500 scientific articles and the gold standard for IP of GFP.

Specificity: GFP, EGFP, CFP, YFP, BFP and many more GFP derivatives.


Description: RFP-Trap is an anti-RFP VHH coupled to agarose or magnetic agarose beads. RFP-Trap or RFP-beads are an affinity resin for immunoprecipitation and pull-down of many red fluorescent proteins. The RFP-Trap is referenced in about 100 scientific articles.

The anti-RFP VHH is also known as RFP binding protein, RFP Nanobody or anti-RFP single domain antibody (anti-RFP sdAb) fragment.

Specificity: mCherry, mRFP, mRFPruby, mPlum, tagRFP, mKate2 and many more RFP derivatives.

The ChromoTek Spot-System is the first peptide-tag specific Nanobody for universal capture & detection applications. It comprises Spot-Tag®, an inert 12 amino acid peptide-tag (PDRVRAVSHWSS), and the universal, rugged Spot-Nanobody that specifically binds to Spot-tagged proteins with high affinity.

Nano-Boosters & Nano-Labels

ChromoTek’s Nano-Boosters & Nano-Labels are fluorescent probes. They consist of very small recombinant antibody domains, also called Nanobodies or VHHs. These single-domain alpaca antibody fragments are covalently coupled to fluorescent dyes.

ChromoTek’s unconjugated Nanobodies or VHHs are well characterized and validated for the listed applications. Because the molecular weight of these single-domain antibodies (sdAb) is just about 15 kDa, VHHs are only one tenth the size of a conventional IgG antibody (150 kDa) or less than a third the size of a Fab fragment (50 kDa). In combination with their high chemical and thermal stability plus high affinity, these binding proteins are ideal for multiple types of affinity assays in biochemistry and microscopy, including immunoprecipitation, immunofluorescence, super resolution microscopy, surface plasmon resonance (SPR), biolayer interferometry (BLI), switchSENSE®* technology, etc.

In addition ChromoTek offer a range of conventional antibodies. These mouse, rabbit, and rat antibodies are used for the reliable detection of fluorescent proteins, protein-tags, and small epitope tags as well as important epigenetic markers, and RNA-Polymerase II.

The conventional IgG antibodies complement the ChromoTek alpaca single domain antibody products due to a different antigen recognition mode: Typically, Nanobodies tend to bind to conformational 3D epitopes, whereas conventional antibodies mostly bind linear epitopes, which can be found in denatured proteins, e.g. on a Western blot.

Chromobodies – Intracellular Functional Antibodies or Intrabodies

ChromoTek’s Chromobodies® are fluorescent nanoprobes for real-time and live-cell imaging of intracellular endogenous proteins.
A Chromobody is a small intracellular functional antibody. It consists of a VHH/ Nanobody that is genetically fused to a fluorescent protein. Unlike conventional antibodies, Chromobodies visualize endogenous cellular structures and processes in live cells and real-time.


Nano-Secondaries are the next level of secondary antibodies. They are based on monoclonal recombinant VHHs/ Nanobodies. Nano-Secondaries bind to primary antibodies with high affinity and specificity. Our Nano-Secondaries are conjugated to Alexa Fluor® dyes. Currently, Nano-Secondaries against rabbit and mouse IgG are available.

Spot-Tag Vectors
